      目的 探讨热休克蛋白70(HSP70)在内毒素血症 大鼠肺损伤中的作用及可能机制。 方法大鼠尾静脉注射脂多糖(lipopolysaccharide,LPS)复制内毒素血症模型。雌性Wistar 大鼠24只,随机分为4组:对照组(C组);内毒素血症组(E组);Gln提前干预组(G1组 );Gln延迟干预组(G2组)。所有的动物于注射LPS之后6 h放血处死,测定肺组织HSP70 的表达、超氧化物歧化酶(SOD)活性及丙二醛(MDA)的含量,并用光镜观察大鼠肺组织的 病理变化。 结果  E组同C组相比,肺组织HSP70表达明显增多(P<0.01),SOD活性明显降低(P<0.01 ),MDA含量增高(P<0.05)。与E组相比,G1,G2组肺组织HSP70表达增多(P< 0.05),SOD活性增高(P<0.05),MDA含量明显降低(P<0.05)。G1与G2在肺 组织HSP70表达,SOD活性,MDA含量等方面比较均无显著性差异(P>0.05)。 结论 增加HSP70的表达对内毒素血症大鼠肺损伤可能起保护作用,作用机制可能与增加肺组织HSP 70,提高机体应激时抗氧化能力有关,尚不能认为Gln提前干预与Gln延迟干预之间有差异, 需要作进一步研究。

      Objective To explore the ef fect and possible mechanism of HSP70 on lung injury in rats with endotoxemia.  Methods  The rat model of endotoxemia was established with injecting lipopolysaccharide(L PS) through caudal vein. Twenty four Wistar female rats were randomly divided i nto four groups: control group(C group),endotoxemia group(E group), Gln pretreated group(G1 group), Gln post treated group(G2 group). All rats were k illed at hour 6 after injecting LPS. Expression of HSP70,the activity of SOD and the contents of MDA in lung tissues were determined. The pathological changes of lung tissue were observed by light microscope. Results Compared with C group, HSP70 expression and the contents of MDA i n the lung tissues significantly increased in E group (P<0.01),the activity of SOD decreased(P<0.01). Compared with E group,HSP70 expression and the ac tivity of SOD significantly increased in G1 and G2 groups (P<0.05) , the contents of MDA decreased(P<0.05),but there was no significant dif ference between G1 and G2 groups(P>0.05). Conclusion The higher expression of HSP70 may protect lung injury in rats with endoto xemia, which may be related to th e high HSP70 induced increase of anti oxidation ability. But it could not be concluded that there is significant difference between Gln pretreatment and pos t treatment. 
