      目的 研究多囊卵巢综合征(PCOS)大鼠雄激素、 胰岛素抵抗及瘦素之间的相互关系。 方法 应用脱氢表雄酮(DHEA)注射法诱导23日龄雌性SD大鼠,建立PC OS动物模型。实验分PCOS组及对照组,每组各15只。采用放射免疫法测定大鼠血清胰岛素( INS)、瘦素(leptin)、卵泡刺激素(FSH)、黄体生成素(LH)、雌二醇(E2)、睾酮 (T)、孕酮(P)水平;采用葡萄糖氧化酶法测定血清血糖(GS)水平。 结果 两组间雄激素水平存在显著性差异(P<0.05),胰岛素释放 试验及葡萄糖耐量试验两组间各时点也均存在显著性差异(P<0.01),PCOS组雄激素 与空腹胰岛素之间呈高度正相关(r=0.889)。PCOS组瘦素与空腹胰岛素、空腹血糖 呈高度正相关(r分别为0.945,0.875)。 结论 PCOS的形成和发展主要与高雄激素血症及胰岛素抵抗相关,而PCO S大鼠血清瘦素水平的升高仅为PCOS大鼠肥胖后的一种伴随现象,瘦素可能通过胰岛素抵抗 作用来调节生殖功能。
英文摘要:
      Objective To study the relationship between androgen, insulin resistance and leptin in rats with polycystic ovary s yndrome(PCOS). Methods The 23 day old female SD rats were injected with droepiand rosterone(DHEA) to establi sh PCOS animal model by. Thirty rats were divided into PCOS group and control gr oup(n=15 in each group). Serum insulin, leptin, follicle stimulin hormone, luteinizing hormone, estradiol, testosterone, progesterone were determined by rad ioimmunoassay. Serum sugar level was determined by glucose oxidase assay. Results The level of androgen had significant difference betw een 2 groups(P<0.05). There were significant difference in oral gluco se tolerance test (P<0.01)and insulin releasing test (P<0.001) at each point time between 2 groups. The androgen was positively correlated with the em pty stomach insulin in PCOS group(r=0.889). Leptin was positively related to emp ty stomach insulin (r=0.945) and empty stomach sugar (r=0.875) in PCOS group. Conclusion Formation and development of PCOS are mainly relat ed to the hyper androgenemia and insulin resistance, whereas the increase of l eptin is a concomitant phenomenon of obese. The results suggest that l eptin may regulate the reproduction function with its insulin resistance.
