      目的〓 观察不同吸烟时间大鼠支气管上皮细胞、肺泡巨噬细胞和肺组织中γ-谷氨酰半胱氨酸合成酶(γ GCS)和转化生长因子-β1(TGF β1)mRNA及其蛋白的表达水平以及二者的相关性,探讨TGF β1在吸烟所致慢性阻塞性肺疾病(COPD)氧化/抗氧化失衡中的作用。 〓方法〓 自制大鼠实验性被动吸烟装置,建立吸烟引起COPD的动物模型,将40只大鼠随机分为不吸烟组、吸烟1月组、吸烟2月组、吸烟3月组,每组各10只。分别采用免疫组织化学法、免疫细胞化学法和原位杂交半定量技术检测支气管上皮细胞及肺泡巨噬细胞中γ GCSh(重链亚单位)和TGF β1 mRNA及其蛋白的表达水平。逆转录聚合酶链式反应(RT PCR)方法检测肺组织匀浆中γ GCSh和TGF β1 mRNA的表达水平。 〓结果〓 吸烟3月组大鼠出现了肺气肿的病理改变。①吸烟1月、2月和3月组大鼠支气管上皮细胞γ GCSh和TGF β1 mRNA及蛋白表达水平、肺泡巨噬细胞γ GCSh和TGF β1蛋白表达水平明显高于不吸烟组(均P<0.01)。②吸烟1月、2月和3月组大鼠肺组织匀浆中γ GCSh和TGF β1 mRNA表达水平明显高于不吸烟组(均P<0.05)。③吸烟1月组和吸烟3月组大鼠支气管上皮细胞γ GCSh mRNA及其蛋白的表达水平与TGF β1表达水平呈直线正相关,肺泡巨噬细胞γ GCSh蛋白表达水平与TGF β1表达呈直线正相关;吸烟3月组肺组织匀浆中γ GCSh mRNA表达水平与TGF β1表达呈直线正相关。 〓结论〓 随着吸烟时间的延长,支气管肺组织中γ GCS和TGF β1的mRNA及其蛋白表达水平逐渐增高,并且二者呈正相关;提示TGF β1在吸烟所致COPD氧化/抗氧化失衡中可能发挥一定的作用。

      Objective〓 To explore the role of TGF β1 in oxidant/antioxidant imbalance of smoking rats with chronic obstructive pulmonary disease(COPD)by investigating the expression of protein and mRNA of γ glutamylcysteine synthetase(γ GCS)and TGF β1 and their relations. 〓Methods〓 Rat smoking device was made by self.Forty rats were randomized into 4 groups:non smoking group(n=10),one month smoking group(n=10),two month smoking group(n=10),three month smoking group(n=10).The changes of bronchial epithelia and macrophages were compared among 4 groups.Immunohistochemistry,cytochemistry stain and hybridization were used to detect the expression of mRNA and protein of γ GCSh and TGF β1 in bronchial epithelia and alveolar macrophages,respectively.In addition, the mRNA expression of γ GCSh and TGF β1 in pulmonary tissues were examined by RT PCR. 〓Results〓 ①Compared with non smoking group,the protein expression of γ GCSh and TGF β1 in bronchial epithelia and alveolar macrophages in one month smoking group,two month smoking group and three month smoking group increased significantly(P<0.01).②Compared with non smoking group,the mRNA expression of γ GCSh and TGF β1 in lung tissue and in bronchial epithelia in one month smoking group, two month smoking group and three month smoking group increased significantly(P<0.05 or P<0.01).③The expression of γ GCSh mRNA and protein in bronchial epithelia and γ GCSh protein in alveolar macrophages was positively correlated with the level of TGF β1 in one month smoking group and three month smoking group.The expression of γ GCSh mRNA in pulmonary tissue was positively correlated with the level of TGF β1 in three month smoking group. 〓Conclusion〓 The results indicate that TGF β1 may play a great role in oxidant/antioxidant imbalance in smoking induced COPD.
