      目的 观察急性坏死性胰腺炎大鼠枯否细胞的分泌功能。方法 将27只健康雌 性Wistar大鼠随机分为三组:假手术组(SO组)、急性坏死性胰腺炎组(SAP组)及三氯化 钆组(GdCl3组)。各组动物均于术后24 h处死。HE染色,光镜下观察胰腺病理改变;检 测 血中内毒素(endotoxin, ET)、淀粉酶(AMS)、肿瘤坏死因子 α(TNF α)、一氧化 氮(NO)及丙二醛(MDA)水平。 结果 SAP组大鼠符合急性出血坏死性 胰腺炎表现;GdCl3组与SAP组相比胰腺病变未见明显减轻。SAP组与SO组相比,血中ET、A MS、TNF α、NO及MDA水平均显著升高(P<0.01);GdCl3组与SAP组相比,血中ET水 平显著升高(P<0.01),AMS水平无明显变化(P>0.05),TNF α、NO及MDA水平 均显著降低(P值分别<0.01,0.05,0.01)。 结论 SAP时血中ET水 平显著升高,枯否细胞(Kupffer cell, KC)分泌功能增强;用GdCl3预处理后血中ET水 平更高,但能减少细胞因子、炎性介质及自由基等的释放。

      Objective To study the secretion function of Kupffer cells in severe acute pancreatitis ra ts.Methods Twenty seven healthy female Wistar rats were ran domly divided into three groups: sham operation group (SO group), severe acute pancreatitis group (SAP group) and gadolinium chloride group (GdCl3 group). Al l rats were killed at hour 24 after operation. The pathological changes of pancrea s were observed by HE staining assisted with light microscope. The blood endoto xin (ET), amylase (AMS), tumor necrosis factor α(TNF α), nitrogen monoxide (NO) and malondialdehyde (MDA) were detected.Results In SAP group, massive necrosis, h aemorrhagia and inflammatory infiltration were found in the pancreas. In GdCl 3 group, the pathological changes were simil ar to the rats in SAP group, which were not significantly lightened. Compared wi th SO group, the levels of blood ET, AMS, TNF α, NO and MDA were significantly higher in SAP group (P<0.01). Compared with SAP group, the level of blood E T was significantly higher in GdCl3 group (P<0.01), the levels of blood TN F α, NO and MDA  significantly decreased (P<0.01, 0.05 and 0.01), but the level of blood AMS had no significant difference (P>0.05). Conclusion In SAP rats, blood ET significantly increases,and the secretion function of Kup ffer cells is enhanced. After GdCl3 pretreatment, blood ET is significantly higher than in SAP group, but the production of cytokine, inflammatory mediator and free radical decreases.
