      目的〓 观察可溶性速殖子抗原(soluble tachyzoite antigen,STAg)于妊娠前不同时间滴鼻免疫小鼠对弓形虫垂直传播的阻断作用。 〓方法〓 BALB/c雌鼠130只随机分为STAg和PBS组,分别用STAg(20 μg/只)和PBS滴鼻免疫2次(间隔2周)。末次滴鼻后雌雄鼠以2∶〖KG-*2〗1分批交配,各组获得孕鼠30只,各组按末次免疫时间的不同(妊娠前3 d, 9 d,15 d)分为3小组,分别为PBS3、PBS9、PBS15组和STAg3、STAg9、STAg15组。全部孕鼠于妊娠第8天用8 000个/只速殖子灌胃攻击。妊娠第18天颈椎脱臼处死,记录孕鼠感染率、活胎率,检测孕鼠小肠冲洗液sIgA水平,测定孕鼠肝、胎盘和胚胎脑组织弓形虫抗原。 〓结果〓 随着免疫时间推迟,STAg组感染率降低,STAg15组感染率最低,但三组间比较无统计学差异(P>0.05),STAg各组孕鼠感染率均低于相应PBS各组,但差异无统计学意义(P>0.05)。STAg组小肠冲洗液sIgA水平逐渐升高,STAg15组高于STAg3组(P<0.05)和STAg9组(P>0.05),STAg15组明显高于PBS各组(P<0.05)。随着免疫时间推迟STAg组活胎率增加,STAg15组活胎率高于STAg3组和STAg9组,但差异无统计学意义(P>0.05),STAg15组活胎率显著高于PBS15组(P<0.05)。STAg组胎盘感染率和胚胎感染率均呈减少趋势(P>0.05),STAg组胎盘和胚胎感染率均低于PBS各组,但差异无统计学意义(P>0.05)。各组胎盘感染率高于胚胎感染率,但差异无统计学意义(P>0.05)。 〓结论〓 妊娠前15 d STAg末次滴鼻免疫小鼠,可有效诱导孕鼠黏膜及系统免疫应答,提高孕鼠抗弓形虫感染作用,降低胎盘、胚胎感染率,减少垂直传播的发生,部分阻断垂直传播。

      Objective〓 To explore the effects of intranasal immunization with STAg at different times before pregnancy on vertical transmission of Toxoplasma gondii in BALB/c mice. 〓Methods〓 BALB/c female mice were randomly divided into STAg group and PBS group.Mice were immunized intranasally with STAg 20 μg and PBS 20 μl per mouse twice at an internal of 2 weeks,respectively.Female mice were mated with males after the last immunization,and 30 pregnant mice were obtained at last in each group.According to the different last immune time before pregnancy,the mice in each group were divided into 3 groups:3 d group,9 d group,and 15 d group.The mice were orally infected with 8 000 tachyzoites of T.gondii RH strain at day 8 after pregnancy.At day 18 after pregnancy,the mice were killed,and the infection rate and average embryo survival rate were determined.The specific sIgA antibodies in intestinal washes and the Toxoplasma antigen in livers,placentas and fetuses of pregnant mice were detected by ELISA. 〓Results〓 The infection rate in STAg3,STAg9,STAg15 groups were lower than that in PBS3,PBS9,PBS15 groups,respectively,but there was no significant difference (P>0.05).The infection rate in STAg15 group was the lowest in STAg groups, but there was no significant difference (P>0.05).The level of sIgA in intestinal washes of pregnant mice increased after immunity.The level of sIgA in STAg15 group was higher than that in STAg3 group(P<0.05)and STAg9 group(P>0.05),and significantly higher than that in PBS groups(P<0.01).〖JP2〗The average embryo survival rate increased gradually in STAg groups.〖JP〗The average embryo survival rate in STAg15 group was significantly higher than that in PBS15 group(P<0.05),also higher than that in STAg3 and STAg9,but there was no significant difference(P>0.05).The placentas and embryo infectious rates in STAg groups were lower than those in PBS groups, but there was no significant difference(P>0.05).And the placentas infectious rate in each group was higher than the embryo infectious rate,but there was no significant difference(P>0.05). 〓Conclusion〓 Intranasal last immunization with STAg at day 15 before pregnancy can effectively induce the mucosal and systemic immune responses in BALB/c mice and protect pregnant mice against Toxoplasma gondii,and mice can be induced to be against the vertical transmission of Toxoplasma gondii by decreasing infectious rates of pregnant mice and the placentas and embryo.
